Antioxidant and Antibacterial Effect of Vitis labrusca, Vitis vinifera and Vitis vinifera Seeds Extract

Grape seeds extract has therapeutic values including antimicrobial activity, antioxidant effect, wound healing and prevention of cardiovascular diseases. This study aimed to evaluate and compare antibacterial activity of different species of grape seed) Vitis labrusca, Vitis vinifera and Vitis vinifera( against some bacterial strains (Staphylococcus aureus, Streptococcus pneumonia, Acinetobacter Calcoaceticus, Klebsiella pneumoniae and Escherichia coli). Determine antioxidant effect of grape seed extracts (qualitatively). Antibacterial effects was performed using agar cup cut diffusion method for all bacterial species, followed by using minimum inhibitory concentration MIC for the species showed to be inhibited by grape seeds extracts. Antioxidant assay was done using DPPH scavenging test, methanolic solution of each grape seeds was spotted on TLC paper, sprayed with 0.2 % methanolic solution of diphenyl picryl hydrazyl (DPPH) reagent. Vitamin C was used as positive control. From the results, all grape species didn’t have any effect on K. pneumonia and E. coli, red and black grape seeds showed the highest inhibition zone (20 mm) on Staph. Aureus agar plate, green grape had the highest effect on Sterp. Pneumonia agar plate (20 mm). The lowest effect was for the red grape seeds extract (13 mm) on Acinetobacter calciaceticus. In general the three grape seeds extract had effect on Staph. aureus, Sterp. pneumonia and Acinetobacter calciaceticus. The red and black grape seed extract was effective against Sterp. pneumonia strain at MIC values of 7.8 mg/mL and black grape seeds extract had MIC at 7.8 mg/mL on Staph. aureus. However, the test for MIC of seeds extracts for the rest of bacterial species ranged between 15.62 and 87.5 mg/mL. The result showed that black grape seeds extract had the largest spot change in color indicating strong antioxidant effect. The lowest effect was by red grape seeds. From this result black grape showed to be the best grape seeds extract among the three chosen species in its antibacterial and antioxidant efficacy.

Phyto-pharmaceuticals and biological study on graviola (Annona muricata L.) fruit and dietary supplement of graviola sold on the Libyan market as a cancer cure against TCA induce hepatotoxicity in mice

Annona muricata (Graviola) has many medicinal properties and used widely in traditional medicine for treatment various disorders. The present study was conducted to evaluate phytochemical and quality control (QC) of random sample of graviola dietary supplement capsules (DS) which sold in the Libyan market as anticancer product. As well as the present work designed to evaluate heatoprotective effects of aqueous extract of graviola fruit pulp or aqueous DS of graviola capsules against trichloroacetic acid (TCA) induced hepatotoxicity in albino mice. Quality control parameters were determined on random samples of graviola DS using standard methods. A total of 120 female mice were divided into 6 groups and were used for biological screening to determine biochemical and histopathological alteration in liver of mice treated with TCA with or without aqueous extract of A.muricata fruit pulp or DS of graviola. The results of quality control and phytochemical screening revealed that all quality control tests conducted on the random sample of DS capsules of graviola were within normal values according to the standards of the Quality Control Center for Pharmaceuticals in Tripoli, only few samples showed slight increase in the moisture content. However, all samples appeared free from microbial contamination. While, growth of fungal contamination (Pencillium Spp) in the same samples were detected but all samples appeared free from aflatoxins contamination. Also, all samples were free from industrial radioactive contamination. Phytochemical study revealed presence of alkaloids, tannins, steroids, glycosides, falvonoids, anthraquinones, saponin and coumarins in extracts of graviola fruit pulp and graviola DS capsules. However, absence some phytochemical components in DS capsules was detected. The result of biological screening revealed that no clinical signs and abnormalities in behavior and external feature in mice treated with aqueous extract of graviola fruit pulp or aqueous extract of graviola DS capsules. However, the treatment with aqueous extract of graviola fruit pulp and DS of graviola reduced the abnormal changes in behavior and external features in female mice intoxicated with TCA, markedly reduced the mortality in TCA administrated mice and induced slight improvement in the final body weight comparing to TCA only intoxicated group. Biochemical study revealed that administration of aqueous extract of graviola fruit pulp or aqueous extract of DS of graviola significantly decreased the elevated serum activities of AST and ALT compared to TCA only intoxicated mice. Histological examination revealed that administration of aqueous extract of graviola fruit plup or aqueous extract of DS of graviola with TCA induced ameliorative changes and disappearance of the most pathological changes in the liver tissue compared to of TCA only intoxicated mice and the ameliorating changes were more obvious in the mice treated with aqueous extracts of DS of graviola and TCA. The present results demonstrate that A. muricata play an important role in the protection against TCA induced hepatotoxicity. It can be concluded that the present study provide some pharmacological and therapeutical informations about extract of the graviola fruit pulp and DS of graviola capsules which can use in future investigations and applications and demonstrated presence of important phyochemical constituents in the graviola fruit pulp extract and DS of graviola capsules. دراسة الكيميانباتية والقيمة الغذائية والتأثير المضاد للتأكسد والمضاد للتخثر لنبات القازول الذي ينمو في منطقة مسلاته – ليبيا" Phytochemical, Antioxidant, Anticoagulant Activities and Nutritional value of Allium roseum L (Alliaceae) Grown in Mesellata region- Libya

نبات القازول Allium roseum L نبات ينتمي إلى الفصيلة البصلية Alliaceae مستوطنا في شمال أفريقيا. يستعمل هذا النبات خلال فصل الربيع في إعداد بعض الوجبات الشعبية كما يستعمل كبديل عن الثوم Allium sativum.يستهدف هذا البحث دراسة الكيمياء النباتية لنبات القازول لمعرفة المواد الفعالة ولمعرفة القيمة الغذائية ومعرفة تأثيره كمضاد للتأكسد وكمضاد للتخثر.خضعت الأزهار والأبصال الطازجة كلا على حدة إلى التقطير البخاري وذلك لاستخلاص الزيوت الطيارة. كما خضعت الأزهار والأبصال المجففة في الظل للاستخلاص عن طريق عملية النقع في الميثانول في درجة حرارة الغرفة.طبقا لطرق التحاليل الكيميائية المرخصة (AOAC 2002) فإن النتائج أثبتت أن النبات يعتبر مصدرا جيدا للبروتين والدهون والألياف والسكريات حيث كانت (13.436 ±0.23, ,5.503 ±0.26 31.032 ±0.0و29.839±0.19) متتابعة بالنسبة للأزهار؛ (19.361 ±0.26, ,14.813 ±0.09 15.803 ±0.06 و34.714±0.9) متتابعة بالنسبة للأبصال.تم التعرف على التركيب الكيميائي للزيوت الطيارة والمستخلص الميثانولي لكل من الأزهار والأبصال عن طريق استخدام جهاز GC-MS. النتائج بينت أن المركب Triacoctane (40.47%) والمركب Isoelemicin (30.27%) كمكونين أساسيين بالنسبة للزيت الطيار للأزهار؛ والمركب 9,12Tetradecadien1ol,acetate,(Z,E) (88.43%) كمكون أساسي بالنسبة للزيت الطيار المستخلص من للأبصال. كما تم تحليل مستخلصات الميثانول بواسطة GC-MS وتم التعرف على مجموعة من المركبات.هذه الدراسة كشفت أن النبات له تأثير معتدل ضد التأكسد مقارنة بفيتامين ج؛ وأن معدل التأثير المضاد للتأكسد بالنسبة للأزهار أكبر من الأبصال.كما تم دراسة التأثير الخارجي المضاد للتجلط المعروف باسم (وقت البروثرومبين) لمستخلصي الميثانول بأحجام مختلفة (25, 50 و75 ميكروليتر) حيث اختبرت على عينات دم لأشخاص طبيعيين بواسطة قياس وقت البروثرومبين. حيث وجد أنه يزداد الوقت المطلوب للتجلط بازدياد الحجم المضاف من المستخلص وأن مستخلص الأزهار له فعالية أقوى من مستخلص الأبصال .Abstract: This study deals with the chemical characterization of a medicinal and an aromatic plant of the Libyan flora: Allium roseum L, and aimed to identify new bioactive natural compounds in its flowers and bulbs, determine the nutrient and bioactive compounds and screening for their possible antioxidant and anticoagulant properties. The chemical composition of essential oils steam-distillized as well as methanolic extracts of both flowers and bulbs were analyzed by GC/MS. The results showed that the Triacoctane and Isoelemicin were detected as the major components of the flowers essential oil (40.47, 30.27%) repectively. For bulbs essential oil showed that 9,12Tetradecadien1ol, acetate, (Z, E) was detected as the major component (88.43%). The GC-MS analysis of the methanolic extracts of the flowers and bulbs of A. roseum showed the presence of: five compounds in flowers methanolic extract, in terms of percentage amounts 10,13-Octadecadienoic acid, methyl ester (19.92%), 2,2-dimethoxybutane(17.55%), Furan,tetrahydro-2,5-dimethyl (12.02 %) and Hexadecanoic acid, methyl ester (11.68 %); and nineteen compounds in methanolic extract of bulbs, in terms of percentage amounts: Heptadecane (13.96%), Octadecane (10.84%), Tetradeca-(9z,12e)-dienyl acetate (10.47%) and Hexadecane (8.97 %)were predominant in the extract. Association of the official analytical chemists methods (2002) are used for nutritional analysis of flowers and bulbs of A. roseum. Results indicated that this plant is an excellent source ofproteins, fats, fibers and carbohydrates (13.436 ±0.23, 5.503 ±0.26, 31.032 ±0.04 and 29.839±0.19) respectively for flowers and (19.361 ±0.26, 14.813 ±0.09, 15.803 ±0.06 and 34.714±0.9) respectively for bulbs. The study revealed that, A.roseum L had moderate antioxidant, scavenging effect of methanolic extracts of flowers and bulbs when compared with standards (ascorbic acid and quercetin). IC50 (µg/ml) 5 min following the order: quercetin (1.08) < ascorbic acid (2.14) < flower extract (116.17) < bulb extract. Free radical scavenging activity of these samples also increased with an increasing concentration. In vitro anticoagulant effects of methanolic extracts of both flowers and bulbs of A. roseum in different volumes (25, 50 and 75 μL) were examined in the blood samples of normal individuals by measuring prothrombin time (PT). The methanolic extracts of flowers and bulbs of A. roseum were found to inhibit coagulation process in vitro and significantly prolonged prothrombin time in a dose-dependent manner. Methanolic extract of flowers was more potent in prothrombin time prolongation than methanolic extract of bulbs. All results in this project were investigated for the first time on Libyan A. roseum L.
